ER Staining Kit - Red Fluorescence - Cytopainter (ab139482) for endoplasmic reticulum staining. Live HeLa cells stained with Red Detection Reagent (A), Hoechst dye (B) and resulting composite image (C). The ER staining kit (ab139482) is specifically designed for use with GFP-expressing cell lines, as well as cells expressing blue, cyan or yellow fluorescent proteins (BFPs, CFPs, YFPs). Additionally, a nuclear counterstain (Hoechst 33342) is also included in the kit.

F-actin Staining Kit - Blue Fluorescence - Cytopainter (ab112124) for actin filaments staining. Embedded mouse embryonic stem cell-differentiated embryoid bodies (EBs) frozen sections are stained with Rabbit anti laminin alpha 1, Goat Anti-Rabbit IgG H&L (Cy5®) preadsorbed secondary antibody (ab97077) and F-actin Staining Kit - Blue Fluorescence - Cytopainter (ab112124). Immunocytochemistry/ Immunofluorescence - DRAQ5™ (ab108410). HeLa cells were stained with Anti-beta Catenin antibody (ab16051) and Anti-alpha Tubulin antibody [DM1A] - Loading Control (ab7291). The secondary antibodies were Goat Anti-Rabbit IgG H&L (DyLight® 488) preadsorbed (ab96899) and Goat Anti-Mouse IgG H&L (DyLight® 594) preadsorbed (ab96881). DRAQ5™ (ab108410) was added to the secondary antibody mixture to label nuclear DNA (pseudocolor orange). Image key: green (beta Catenin), red (alpha Tubulin), orange (nuclei DRAQ5™).

Immunocytochemistry/ Immunofluorescence - EdU Assay / EdU Staining Proliferation Kit (iFluor 488) (ab219801). EdU Staining Proliferation Kit (iFluor 488) (ab219801) staining of proliferating cells. HeLa (Human epithelial cell line from cervix adenocarcinoma) cells (4 x 10⁴ cells/well in 96 plate) were incubated with 10 μM EdU for 3 hours. Cells were analyzed using a TCS SP8 confocal microscope (Leica-Microsystems). DNA (blue) was staining with Hoechst 33342 (ab145597). Green cells show EdU/Hoechst-positive cells.

粒線體膜電位 (mitochondrial membrane potential) 下降是細胞凋亡 (apoptosis) 的指標之一，同時也是調控細胞凋亡的重要關鍵。已有多項研究證實，癌細胞往往具有異常高的粒線體膜電位，透過降低癌細胞粒線體膜電位，可促使癌細胞走向細胞凋亡 [1]。Abcam 提供多款活細胞分析用螢光染劑產品，可協助您精確檢測藥物處理後的細胞粒線體膜電位變化。

Mitochondrial membrane potential was measured using TMRE-Mitochondrial Membrane Potential Assay Kit (ab113852). P19 neurons (750 cells/mm²) were exposed to MDMA on days 7–9 in serum-free medium for 10 min up to 48 hours. The positive control FCCP (carbonyl cyanide-p-trifluoromethoxyphenylhydrazone), an uncoupler of mitochondrial oxidative phosphorylation, was applied at the concentration of 5 μM for 10 min. The cells were incubated with 500 μM TMRE for 30–45 min at 37°C, 5% CO₂, followed by washing once with 100 μl of HBSS containing 0.2% bovine serum albumin. A volume of 200 μl of HBSS containing 0.2% bovine serum albumin was added to each well, and the fluorescence was measured with excitation/emission: 544/590 nm. Fluo-8 vs Fluo-4 sensitivity to calcium release in HEK-293 cells induced by Carbachol. Carbachol Dose Response was measured in HEK293 cells with Fluo-8 Calcium Flux Assay Kit - No Wash (ab112129) and a Fluo-4 calcium assay kit. HEK293 cells were seeded overnight at 40,000 cells/100 µl/well in a black wall/clear bottom 96-well plate. The cells were incubated with 100 µl of dye-loading solution using the ab112129 or the Fluo-4 kit for 1 hour at room temperature. Carbachol (50 µl/well) was added to achieve the final concentrations indicated.

ATP Dose Response in CHO cells measured with Fura-2 Calcium Flux Assay Kit - No Wash, Ratiometric (ab176766). CHO-K1 cells were seeded overnight at 40,000 cells/100 μl/well in a Costar black wall/clear bottom 96-well plate. The cells were incubated with 100 μl of the Fura-2 Calcium Flux Assay Kit - No Wash, Ratiometric (ab176766) for 1 hour at room temperature. ATP (50 μl/well) was added to achieve the final indicated concentrations.
