SPA 技術
SPA (Scintillation Proximity Assay) 鄰近閃爍分析技術,是由 GE Healthcare 研究團隊所研發建立的全均質化放射性偵測平台,應用領域包含 Molecular Interaction、Receptor-Ligand Binding Assay、Enzyme Activity Assay、Biomolecule Detection、Cell Proliferation、Cell Adhesion 等…多種分析實驗。至目前為止已具有超過 500 篇以上的文獻發表紀錄,被視為是放射性大量篩選試驗的 "Gold Standard" 技術。

SPA 技術原理
SPA Beads (或 Cytostar-T Scintillating Microplate 盤底) 材質內含閃爍計數劑,當帶有 β 射線核種 (如:3H, 14C, 33P, 125I) 的生物分子藉由親合性結合或細胞攝入的方式,靠近 SPA Beads (或 Cytostar-T Scintillating Microplates 盤底) 時,即可藉由能量轉移使放射線轉換為光能。而未能與 SPA Beads 結合 (或未能被 Cytostar-T Scintillating Microplates 盤子底部的細胞攝入) 的游離放射線物質,則會因為距離過遠而無法誘使其產生光訊號。研究者可藉由光值訊號的強弱,了解分子結合作用或細胞生長的情形。 |

SPA Beads
由鄰近閃爍分析技術所延伸出來的便利產品,微珠本身含有閃爍計數劑成分,適用於多種放射線偵測實驗,例如受體結合、磷酸酶活性、分子交互作用等。全均質化的實驗操作步驟,除了可讓實驗者省略麻煩的分離洗滌步驟之外,更可節省購買閃爍計數液或濾盤的費用。依據最終發光訊號波長的不同,產品可區分為適合以 CCD 影像系統偵測的 SPA Imaging Beads,以及適合以盤式輻射偵測儀偵測的 SPA Scintillation Beads。兩者皆有多種規格可供選擇,歡迎洽詢索取詳細技術資料。 |

Cytostar-T 放射線閃爍計數盤為白色盤身透明底部的微量多孔盤,盤底材質內含閃爍計數劑成份,適合貼式細胞的培養、影像觀察與放射性活細胞分析試驗。
Cytostar-T 放射線閃爍計數盤的偵測原理為鄰近閃爍分析法 (Scintillation Proximity Assay, SPA)。以常見的細胞增生實驗為例,當帶有放射線標定的 Thymidine 被盤子底部的細胞攝入時,由於此時放射線靠近盤底,因此可與盤底材質中的閃爍計數劑產生能量轉移反應,使放射線轉換為光能。而未被細胞攝入游離於培養液中的放射線物質,則會因為無法靠近盤底因此無法產生光值訊號。藉由偵測光訊號強度,即可評估細胞生長狀態。
